Phenotypic and genotypic characterization of influenza virus mutants selected with the sialidase fusion protein DAS181.

BACKGROUND influenza viruses (IFVs) frequently achieve resistance to antiviral drugs, necessitating the development of compounds with novel mechanisms of action. DAS181 (Fludase), a sialidase fusion protein, may have a reduced potential for generating drug resistance due to its novel host-targeting mechanism of action. METHODS IFV strains B/Maryland/1/59 and A/Victoria/3/75 (H3N2) were subjec...

Structures of NS5 Methyltransferase from Zika Virus

The Zika virus (ZIKV) poses a major public health emergency. To aid in the development of antivirals, we present two high-resolution crystal structures of the ZIKV NS5 methyltransferase: one bound to S-adenosylmethionine (SAM) and the other bound to SAM and 7-methyl guanosine diphosphate (7-MeGpp). We identify features of ZIKV NS5 methyltransferase that lend to structure-based antiviral drug di...

West Nile Virus Methyltransferase Domain Interacts with Protein Kinase

Background: The flaviviral nonstructural protein 5 (NS5) is a phosphoprotein, though the precise identities and roles of many specific phosphorylations remain unknown. Protein kinase G (PKG), a cGMP-dependent protein kinase, has previously been shown to phosphorylate dengue virus NS5. Methods: We used mass spectrometry to specifically identify NS5 phosphosites. Co-immunoprecipitation assays wer...

Discovery and molecular characterization of West Nile virus NY 1999.

The field of infectious diseases is enjoying a renaissance in professional and popular media. This interst derives primarily from three concerns: bioterrorism, antibiotic resistance, and emerging infectious diseases. In this article we only discuss the last of these; however, it is important to recognize that the public health infrastructure required to address each is similar. The 1999 outbrea...

West Nile Virus, Guadeloupe

To determine whether West Nile virus (WNV) had reached the archipelago of Guadeloupe, a serologic study in horses and birds was conducted in 2002. Immunoglobulin (Ig) G, IgM, enzyme-linked immunosorbent assay, and seroneutralization tests identified WNV infection in horses and chickens. Six months later, a high rate of seroconversion was observed in horses.
